Studies on the Sampling, Fixing, Staining and Reading in the Vaginal Smear
Chung Byung-Ho

Abstract
In the making smear preparation which is chiefly used in the fields of Ob. & Gy., 97 cases of normal & abnormal vaginal smear preparations were made and Papanicolaou¢¥s procedure was discussed and considered as a clinician.
1. In collecting examined specimen, a blunt of, my own style, small curette or cotton applicator applied and dried with collodion U.S.P. were used for scraping and wiping and relatively fresh clear slides were obtained in detecting cancer cells and studing hormonal effects.
2. Positive ratio is relatively high even in the using routine fixation solution such as 95 per cent ethylalchol, acetone and 99 per cent methyl alcohol.
3. Proper fixation time is between 1-2weeks without regarding to the staining of nucleus and cytoplasm } of cell.
4. Cytological method is prior to biopsy procedure in proper control cf the procedure cyto-diagnosis was obtained rapidly by using simple staining which is subsided bacteriological staining or tissue staining.
5. Observing by about microscopic finding of cells and nuclei prior to tissue examination or observing both of them, at the same time, in compare with Papanicolaou¢¥s technique, by examiner who had well trained and made the sampling, fixation, staining, and reading of specimen, my procedure is more simple, proper technique and cheep in price, less suffering to patients, at the same time, morphological changes in individual cell could be observed in short time (10-30-60 minutes). I¢¥m inclined to think that my procedure is simple and easy to perform to everyone who desires to do it at office, clinic, operating room or screening test or anywhereelse.
